My Win 7 Ultimate PC doesn't connect pendrives anymore. I clicked on devmgmt.msc and checked it out, and it simply shows USB Mass Storage Device with a yellow '!' beside it. Tried 'update Driver software', still no progress. Any advice?

Uninstall the driver and restart your PC. All uninstalled drivers will be automatically re-installed. If the problem still persists, change the hardware.

If you used that pendrive to make a booteable drive, try using a partition manager software and "create a partition table" on that drive, then create a partition with full size.

Windows is stupid on anything related to filesystems.

Did you update BIOS or CHIPSET/Motherboard firmware? Sometimes it could be related to the two.
